摘要
目的检测3种白血病细胞Jurkat、Molt-4、HL-60的雌激素受体α(estrogen receptorα,ERα)基因启动子ERα-A、ER-αB CpG岛甲基化状态,初步探讨ERα启动子区的甲基化状态以及与白血病发病机制的可能联系。方法应用甲基化特异性PCR技术(MSP)检测3种白血病细胞株与正常人外周血单个核细胞ERα启动子区ERα-A和ERα-B启动子区CpG岛甲基化状态。结果白血病细胞株ERα-A和ERα-B均出现甲基化条带,为全甲基化状态;正常人外周血单个核细胞ER-αA和ER-αB CpG岛呈现非甲基化状态。结论ERα基因启动子ERα-A和ER-αB CpG岛的甲基化状态有可能成为白血病新的分子诊断标记物。
Objective To investigate the methylation status of CpG islands of the Estrogen receptor (ERα) gene promoter in Jurkat,Molt-4,HL-60,and try to find the possible relationship and pathogenic mechanism of leukemia. Methods The methylation status of CpG island of ERα(ERα-A and ERα-B ) gene promoter in 3 leukemic cell lines and 2 normal white blood cells were studied by methylation specific PCR(MSP). Results ERα A and ERα B promotors were methylatied in 3 leukemic cell lines; 2 normal white blood cells showed unmethylation bands. Conclusion The methylation status of CpG island of ERα(ERα-A and ERα-B ) gene promoters may be a potential indepengdent molecular diagnostic factor of leukemia.
