摘要
目的:观察辛伐他汀对高脂血症大鼠脂肪细胞凋亡的影响,探讨辛伐他汀降低血脂的分子生物学机制,为临床应用提供理论和实验依据。方法:采用自制高脂饲料制备高脂血症大鼠模型,观察大鼠血脂指标以及脂肪细胞凋亡指数的变化。结果:高脂血症组大鼠血清TG、Tch及VLDL-ch含量均显著高于正常对照组(P<0.01),而辛伐他汀组大鼠血清TG、Tch及VLDL-ch水平较高脂血症组均显著下降(P<0.01),高脂血症组大鼠血清HDL-ch水平均显著低于正常对照组(P<0.01),辛伐他汀组大鼠血清HDL-ch较高脂血症组显著增高(P<0.01);且接近正常组水平。高脂血症组大鼠脂肪细胞凋亡指数均显著低于正常对照组(P<0.01),而辛伐他汀组大鼠脂肪细胞凋亡指数较高脂血症组均显著升高(P<0.01)。结论:辛伐他汀降低高脂血症大鼠血脂,促进脂肪细胞凋亡,从而达到治疗作用。
Objective:Through investigating the effect of simvastatin on lipocyte apoptosis in hyperlipoidemia rat,we researched the molecular mechanism of blood lipid reduction function of simvastatin and provided a principle for the practice of simvastatin in clinic. Methods:To prepare the hyperlipoidemia rats by self-preseribed high calorie diet.The change of blood lipid and the index of lipocyte apoptosis were measured.Results:Compared with the control, the level of TG,Tch and VLDL-ch of hyperlipoidemia rat increased obviously (P〈0.01),the level of TG,Tch and VLDL-ch of hyperlipoidemia rat with simvastatin decreased obviously (P〈0.01),and the level of HDL- ch of hyperlipoidemia rat decreased obviously(P〈0.01),the level of HDL-ch of hyperlipoidemia rat with simvastatin increased obviously(P〈 0.01) and the index of lipocyte apoptosis was measured.Compared with the control,the level of the index of lipocyte apoptosis of hyperlipoidemia rat decreased obviously (P〈0.01),while the index of lipocyte apoptosis of hyperlipoidemia rate with simvastatin increased obviously(P〈0.01).Conclusion :During the rats are treated by simvastatin,simvastatin may improve lipocyte apoptosis in order to attain bloodlipid-reduction.
