甘丙肽对嗅成鞘细胞三种神经营养因子mRNA表达的影响

Effect of galanin on mRNA expressions of three neurotrophic factors in olfactory ensheathing cells

摘要 实验从新生大鼠嗅球中分离出嗅成鞘细胞,进行体外培养。运用半定量RT-PCR方法检测甘丙肽对体外培养的嗅成鞘细胞中三种神经营养因子(LIF、CNTF和GDNF)mRNA表达的影响。实验以甘油醛-3-磷酸脱氢酶(G3PDH)作为内参照。结果显示:体外培养的嗅成鞘细胞表达此三种神经营养因子的mRNA;当甘丙肽作用于细胞3d后,嗅成鞘细胞中LIF和GDNF表达量明显降低,而CNTF的表达量则没有发生明显变化。 Olfactory ensheathing cells (OECs) were isolated from newborn rat olfactory bulb and cultured in vitro. Semi-quantitative RT-PCR was used to determining the effect of galanin on mRNA expressions of three neurotrophic factors ( LIF, CNTF and GDNF ) in these cells. The human glyceraldehyde-3-phosphate dehydrogenase (G3PDH) was considered the internal standard. Results showed that OECs expressed mRNAs for the three factors in vitro. Moreover, after OECs were incubated with galanin for 3 days, the expression levels of GDNF and LIF mRNA in drug group were lower than that in control group evidently, but there was no difference of CNTF expression between two groups in these cells.
