摘要
目的:研究成年大鼠嗅鞘细胞(OECs)分离与培养的方法,为进一步进行基础研究以及临床应用提供高纯度的细胞。方法:采用改良差速贴壁法从成年大鼠的嗅球培养出OECs,并用Forskolin和碱性成纤维细胞生长因子(bFGF)促增殖,最后进行OECs特异标记物NGFRp75、S-100的免疫细胞化学染色鉴定OECs。结果:改良差速贴壁法成功培养出高纯度的OECs。结论:此法简便、经济、纯化效率高,Forskolin和bFGF能够促进OECs的增殖。
Aim: To investigate the technical methods for dissociating and primary culturing the olfactory ensheathing cells(OECs)from adult rats, and to provide highly purified cells for the basic and clinical researches. Methods: Purifying the OECs from the olfactory bulbs of adult rat according to the differing rates of attachment of the various harvested cells types. Forskolin and bFGF were added into the cultures, stimulating the OECs to divide and enrich. Eventually the OECs were immunostained with NGFRp75 and S-100 antibody to identify. Results : Highly enriched population and highly purified OECs are harvested with the method. Conclusion: The reported method of differing rates of attachment is simple, economic and with high purification efficiency. Forskolin and bFGF are capable of promoting the proliferation of the OECs.
