胃癌胃周淋巴管、血管分布及变化规律 被引量:2

The regularity of lymphatic and blood vessels distribution and their changes in orthotopically tumor-implanted nude mice

摘要 目的探讨胃癌生长过程中胃周淋巴管、小血管分布及变化规律。方法以SGC- 7901胃癌组织块植入裸鼠胃壁,尔后每隔1周处死1批,取胃周组织切片、双酶染色和苏木素-伊红(HE)染色,光镜观察淋巴管及微血管并图像分析,荷瘤组为实验组,与正常裸鼠对照。结果早期荷瘤鼠的淋巴管及微小血管面积、直径、不规则程度、密度与对照组比较无明显变化(P>0.05);随肿瘤生长,胃周淋巴管面积、最大直径、平均直径呈总体增加趋势,植瘤3周后较3周前比较差异有统计学意义(P<0.05),总体荷瘤鼠淋巴管密度比正常裸鼠高,差异有统计学意义(P<0.05);胃周血管面积、最大直径、平均直径、不规则程度随肿瘤生长呈总体增高趋势,植瘤2周后较2周前比较差异有统计学意义(P<0.05),密度变化差异无统计学意义(P>0.05);原位植瘤6周后可见胃周5’-核苷酸酶(5’-Nase)活性管道与碱性磷酸酶(ALPase)活性管道存在相通现象。结论随肿瘤生长胃周组织淋巴管密度增加、管径增粗;血管变化以管径增粗为主。晚期移植瘤的胃周血管可能与淋巴管存在相通现象。 Objective To investigate the regularity of lymphatic and blood vessel distribution and their changes in orthotopically tumor-implanted in different stages. Methods SGC-7901 tissue blocks were implanted into the anterior wall of stomach of nude mice and one group were kiUed every week after operation. The whole epigastric organs and connective tissue were removed and cut. The sections were stained with double-enzyme and HE staining, then examined under light microscope and analyzed with the image analysis system. Results There was no significant difference in the data of lymphtics between normal mice (W0) and earlier implanted nude mice ( W1-W3 ). FoUowing the growth of tumors, the area, maximal diameter, minimal diameter and average diameter of lymphatics all tended to be larger. The above data were significantly larger in W4-W9 than in W0-W3. The density in total implanted mice was significantly higher than that in normal mice. No significant difference was found in the data of blood vessels between normal mice (W0) and earlier implanted nude mice (W1 and W2). Area,maximal diameter,minimal diameter,average diameter and roundness of vascular vessels all tended to be higher after orthotopic implantation. The data in W3-W9 were significantly higher than those in WO-W2, but no significant difference was found in the density of blood vessels of nude mice at different stages. Six weeks after implantation,in the connective tissues around stomach, communication between the vessels with 5'-Nase activity and with ALPase activity was found. Conclusion With the growth of orthotopic tumor, the diameter and density of lymphatics in the connective tissues around stomach became higher, but the main change of blood vessels was shown in the increase of area. It was possible that the lymphatics and blood vessels were communicated with each other in nude mice with advanced stage stomach cancer.
