摘要
采用ISSR分子标记技术,对一马氏珠母贝养殖群体中个体大和个体小的两个组的遗传多样性进行分析。5个引物在2个组100个样品中共得到72个清晰的扩增位点,其中多态性位点71个,多态位点百分率(PPB)为98.61%。POPGENE分析结果表明:个体大的组的遗传多样性水平高于(PPB=94.44%,I=0.3523,h=0.2181,na=1.9444)小个体组(PPB=80.61%,I=0.3008,h=0.1915,na=1.8056)。两组间的遗传分化系数Gst=0.0948。UPGMA聚类分析表明大个体组内几乎所有的贝(47个)聚在一起,而小个体组的贝(50个)聚成另一支。
One big group (D group) and one small group (X group) of pearl oyster Pinctada martensii, selecting from the same artificial population, were analyzed by ISSR (inter-simple sequence repeats ) markers to determine the genetic diversity among the groups. A total of 72 loci, including 71 polymorphic loci, were amplified using 5 primers. Polymorphic loci were 98.61% in the two groups. The result of POPGENE analysis indicated that the level of genetic diversity of the big group of Pinctada martensii ( PPB=94.44 %, I=0.352 3, h= 0.218 1, na = 1.944 4 ) was higher than the small group ( PPB=80.61%, I=0.300 8, h= 0.191 5, na =1.805 6 ). A total of Gst ( coefficient of population differentiation ) 0.0948 showed that there was 9.48 % of genetic differentiation between the two groups. The dendrogram of genetic relationships among groups and individuals was constructed based on the Jarrced's genetic distance. The 47 of 50 big individuals clustered together, the 50 small individuals clustered another group.
