摘要
目的:研究顺铂、卡铂上调宫颈癌SiHa细胞株胸苷磷酸化酶(thymidine phosphorylase,TP)的变化,探讨序贯联合化疗时抗嘧啶类药物用药的适合时机。方法:体外培养SiHa细胞株,分为对照组、顺铂组、卡铂组。顺铂和卡铂72h的IC50分别是:19μmol/L,236.82μmol/L。药物作用后0、4、8、12、16、24、36、48h,用RT-PCR检测各组细胞TP mRNA表达。于药物作用后0、6、12、18、24、30h,用Western blot检测TP蛋白表达。结果:顺铂和卡铂作用4—8h后TP mRNA表达开始增加,24h达高峰,相对于空白组,顺铂组升高2.52倍,卡铂组升高1.90倍,24h后逐渐降低;顺铂和卡铂作用6—12h后TP蛋白增加,24h达高峰,顺铂组和卡铂组表达分别是对照组的3.45倍和2.73倍。蛋白质增加幅度大于mRNA,顺铂组与卡铂组差异无统计学意义。结论:顺铂和卡铂可以上调宫颈癌SiHa细胞株TP,渐次升高后下降,呈时间依赖性,顺铂组与卡铂组上调TP的作用差异不显著。
Objective:To study if cisplatin and carboplatin could regulate thymidine phosphorylase expression in cervical cancer lines in vitro, to figure out how to combine pryimidine and sequential chemotherapy. Methods:SiHa cell lines were divided into 3 groups:cisplatin group, carboplatin group and the control group. After 72 hours treatment with different drugs,the IC50was 19μ mol/L for cisplatin,236.82μmol/L for carboplatin. TP mRNA expression was detected at 0,4,8,12,16,24,36,48 hours after cisplatin and carboplatin treatment by using semi-quantitative RT-PCR. In addition, TP protein expression was examined at 0,6,12, 18,24 and 36h after drug treatment by using Western blot analysis. Results: TP mRNA expression began to mcreaee 4 - 8 hours after the treatment and reach the maximum at 24 hour. For the protein,the increasing time and peak were located at 6 - 12 hours and 24 hours,respectively. TP mRNA expression in cisplatin group and carboplatin group were 2.52 times and 1.9 times of the control group while the protein expression of cisplatin group and carboplatin group were 3.45 times and 2.73 times that of the control group. TP protein expression increased more significantly than mRNA ,and the difference between carboplatin group and cisplatin group was not statistically significant. Conclusion: Cisplatin and carboplatin can up-regnlate thymidine phosphorylase in SiHa cell line, TP increases gradually and decreases subsequently. There was no difference between cisplatin and carboplatin on the TP regulating in SiHa cell line.
