摘要
胰岛素样生长因子I(IGF-1)是生长激素发挥促生长作用的重要调节因子。本研究为了阐明营养限制和补偿生长对蒙古绵羊IGF-1 mRNA表达水平的影响,选择健康的蒙古绵羊36只,分为0.20MJME/kgw0.75/d(限制组)、0.33MJME/kgw0.75/d(阈值组)、以及自由采食组(对照组)3个营养水平处理组,采用反转录PCR(RT-PCR)技术检测了蒙古绵羊在限制营养后及补偿生长后肝脏和背最长肌中IGF-1 mRNA表达量的变化。结果显示:在限制结束时,限制组母羊肝脏和背最长肌IGF-1mRNA表达量都极显著低于对照组(p<0.01),阈值组显著低于对照组(p<0.05);补偿生长后,限制组和阈值组的IGF-1mRNA的表达量都有所提高,与对照组差异均不显著(p>0.05),说明限制组和阈值组IGF-1mRNA的表达又得到恢复。
IGF-1 is an important adjustive factor to animals growth and development of GH.This study focused on the effects of nutrition restriction and compensatory growth on expression of IGF-1 mRNA of Mongolia sheep.36 Mongolia sheep were selected and divided into three groups offered 0.20 MJME·kgw-0.75·d-1(Restricted Group),0.33 MJME·kgw-0.75·d-1(threshold group) and voluntary intake(Control Group).The expression of IGF-1 mRNA in liver and longissimus dorsi muscle in Mongolia sheep in different nutrition level was examined by RT-PCR.The result indicated that,At the nutrition restriction phase,the abundance of IGF-1 mRNA in liver and longissimus dorsi muscle in Restricted Group was significantly lower than that of the Control Group(p〈0.01),but in threshold group was significant when compared to that of Control Group(p〈0.05);At the ending of the compensatory growth phase,the abundance of IGF-1mRNA in liver and longissimus dorsi muscle in Restricted Group and threshold group were not significant when compared to that of Control Group(p〉0.05).
