摘要
以新牧1号和新疆大叶两个苜蓿品种为试材,研究了7个培养基对不同无菌苗外植体愈伤组织诱导和生长影响,以及不同浓度的植物生长调节物质6-苄氨基腺嘌呤(6-BA)和萘乙酸(NAA)对愈伤组织生长及分化的作用。结果表明:(1)浓度在2-8 mg/L,NAA有利于苜蓿愈伤组织的诱导,随浓度的增大呈促进作用,而在1-3 mg/L,1mg/L 6-BA有利于苜蓿愈伤组织的诱导,随浓度的增大呈抑制作用;(2)MS+2,4-D 2mg/L+NAA 4 mg/L+6-BA 1 mg/L即MS4为新牧1号的较佳诱导培养基,MS+2,4-D 2 mg/L+NAA 8 mg/L+6-BA 1 mg/L即MS5为新疆大叶苜蓿的较佳诱导培养基;(3)以无菌苗下胚轴、子叶及叶片为外植体,确定下胚轴为诱导愈伤组织的最佳外植体。
The paper studied effects of seven kinds of medium on induction and growth of callus, using different of explant of Medicago varia, cv. Xinmu No. 1 and Medicago sativa L. cv. Xinjiang Daye, and effects of different concentration of 6- BA(6- Benzylaminopurine) and NAA( 1 - Naphthyacetic Acid) on induction, dif- ferentiation callus. The results are as follows: ( 1 ) The concentration of 2 - 8 mg/L, NAA is benefit to induce callus, if it's concentration was higher,the callus of alfalfa were better.The concentration of 1 - 3 mg/L, lmg/L 6 -BA is benefit toinduce callus, if it's concentration was higher, the callus of alfalfa were worse. (2)MS + 2,4- D 2 mg/L + NAA 4 mg/L + 6 - BA 1 mg/L was the better induction medium of Xinmu No. 1. MS + 2,4 - D 2 mg/L + NAA 8 mg/L + 6 - BA 1 rng/L was the better induction medium of Xinjiang Daye. (3)Using the explant of hypocotyls, cotyledon and leaves as explants, hypocotyls was the best explant of induction callus medium.
