摘要
目的研究刺五加皂苷(ASS)对血管内皮生长因子(VEGF)蛋白及mRNA表达的影响。方法以人肝癌细胞株(HepG2)为研究对象,分别用ELISA及RT-PCR检测ASS对HepG2细胞VEGF蛋白及mRNA水平表达的影响。结果ASS250,500μg/mL作用于HepG2细胞株,VEGF蛋白及mRNA表达量下降(P<0.05或P<0.01)。结论一定剂量的ASS能抑制HepG2细胞株的VEGF mRNA表达及蛋白的表达,提示ASS可能是通过抑制VEGF介导的肿瘤血管新生,进而抑制肿瘤的生长与转移。
Objective To investigate the effect of Acanthopanax senticosus saponin (ASS) on the protein and mRNA expression of vascular endothelial growth factor (VEGF) in human HepG2. Methods HepG2 were incubated with ASS, the expression of VEGF was detected by ELISA assay, and the mRNA expression of VEGF was detected by RT - PCR as- say. Results ASS (250, 500 μg/mL) decreased the expression of VEGF protein and VEGF mRNA ( P 〈 0.05 or P 〈 0.01). Conclusion The anti - tumor - growth mechanism of ASS is probably related to the inhibition of angiogenesis by decreasing the expression of VEGF protein and VEGF mRNA.
