摘要
目的观察榄香烯乳对视网膜母细胞瘤(Rb)HXO-RB44细胞凋亡抑制蛋白survivin表达及端粒酶活性的影响,探讨榄香烯乳对Rb HXO-RB44细胞的作用机制。方法用不同浓度榄香烯乳作用于HXO-RB44细胞,采用四甲基偶氮唑蓝(MTT)比色法测定细胞增生活性;RT-PCR检测survivin mRNA的表达;聚合酶链反应方法检测端粒酶活性。结果榄香烯乳处理后HXO-RB44细胞受到不同程度的抑制,survivin mRNA表达下降;RB细胞系端粒酶活性减弱。结论榄香烯乳诱导HXO-RB44细胞凋亡与survivin有关;可减弱RB细胞系端粒酶活性,影响其周期变化,抑制RB细胞系增生。
Objective Retinoblastoma is the most common intraocular malignant neoplasm,and it is a big threat to eyesight and lifespan for children. Chemotherapy has become the most important treatment to retinoblastoma in recent years. Elemene is a natural compound that was isolated from the traditional Chinese medicinal herb, Rhizoma zedoarie. We studied the expression of survivin and telomerase activity in human retinoblastoma HRO-RB44 cell treated with elemene and investigated the mechanism of elemene on HXO-RB44 cells. Methods The HXO-RB44 cells were cultured at 37 ℃in 50 mL/L CO2/air for 24,48 and 72 hours and divided into experimental group and control group. The latter was administered by elemene at the concentrations of 20,40,60,80 and 100 μg/mL. Telomerase activity was detected by PCR,and expression of survivn mRNA was detected using RT-PCR. Results The absorbing value of HXO-RB44 and inhibiting rate of elemene were significant reduced in different time points after treated of various concentrations of elemene in comparison with respective control group ( P = 0. 000). Expression of survivin mRNA decreased by 79% ,56% ,43% respectively in various concentrations of elemene groups compared with blank control groups at 24 hours(P 〈 0. 01 ). The telomerase activity had not obvious alteration in different concentrations of elemene groups at 24 hours but was significant different between experimental group and blank control group. Elemene could down-regulate the expression of survivin and decreased telomerase activity. Conclusion Elemene inhibits human retinoblastoma HXO-RB44 cells by down- regulating the expression of survivin and suppressing telomerase activity in a concentration-dependent manner.
