鱿鱼皮胶原蛋白多肽对B16黑素瘤细胞黑素合成的影响 被引量:23

摘要 目的研究不同分子量鱿鱼皮胶原蛋白多肽SP1(Mr>10000u)、SP2(6000u<Mr<10000u)、SP3(2000u<Mr<6000u)对小鼠B16黑素瘤细胞黑素含量、酪氨酸酶活性及酪氨酸酶基因表达的影响。方法采用MTT法测定细胞增殖活性,NaOH裂解法测定黑素含量,多巴氧化法测定酪氨酸酶活性,逆转录-聚合酶链反应(RT-PCR)测定酪氨酸酶mRNA表达。结果不同分子量鱿鱼皮胶原蛋白多肽均能明显降低B16黑素瘤细胞黑素含量(P<0.05,P<0.01),抑制酪氨酸酶活性(P<0.01),并下调酪氨酸酶mRNA表达(P<0.05,P<0.01),且剂量效应关系明显。结论不同分子量鱿鱼皮胶原蛋白多肽均具有明显抑制B16黑素瘤细胞黑素合成的作用,其中SP2的抑制效果较SP1、SP3明显(P<0.05)。 Aim To study the effects of different molecular weight of collagen polypeptides from squid(Dosidicus gigas)skin(SP1:Mr〉10000 u,SP2:6 000 u0.05),the melanogenesis and tyrosinase activity were inhibited remarkably(P〈0.05,P〈0.01).The TYR mRNA level was decreased evidently(P〈0.05,P〈0.01) in a dose dependent manner by SP1,SP2 and SP3,respectively.Conclusion Different molecular weight of collagen polypeptides from squid(Dosidicus gigas)skin can inhibit melanogenesis in B16 melanocytes observably.The inhibition effect of SP2 is better than that of SP1 and SP3(P〈0.05).
