Permeability and Protective Effect of Nerve Growth Factor by Periphery in Neonatal Rats with Hypoxic-Ischemic Brain Damage

摘要 目的探讨外周应用神经生长因子(NGF)能否透过缺氧脑损伤新生鼠血脑脊液屏障及其对脑的保护作用。方法制备缺氧脑损伤新生大鼠模型后,静脉注射125I标记NGF,5μg/kg。给药30min后取脑组织,12h内测定125I放射度,计算NGF水平。同时取血检测其NGF水平。dUTP缺口末端标记法(TUNEL法)观察NGF治疗后新生缺氧大鼠额叶、海马区细胞凋亡情况。结果外周应用125I-NGF后,各组大鼠脑内均可检测出125I-NGF,新生鼠脑组织125I-NGF水平及其与血液中水平的比值显著高于成年鼠(Pa<0.05),新生缺氧鼠脑组织125I-NGF及其与血液中水平比值显著高于正常新生鼠(Pa<0.05)。应用NGF治疗后,NGF治疗组额皮质和海马CA1区凋亡阳性细胞比值低于非NGF治疗组(Pa<0.05)。结论外周应用NGF可通过新生大鼠HIBD血脑脊液屏障,具有一定脑保护作用。  Objective To explore the permeability of nerve growth factor(NGF)through blood brain barrier and protective effect of NGF by periphery in neonatal rats with hypoxic-ischemic brain damage(HIBD).Methods The models with HIBD were established and NGF 5 μg/kg was injected intravenously with ^125Ⅰ,and the permeability of NGF was observed by periphery through blood brain barrier of neonatal rats.Then apoptosis in each group was examined in tissues,samples of frontal cortex and CA1 district in cornu ammonis by TUNEL method.Results The contents of ^125Ⅰ-NGF in neonatal rats and ratio with blood content was higher than those of adult rats,then contents of ^125Ⅰ-NGF in neonatal rats with HIBD and ratio with blood content were higher than those of the neonatal rats(Pa〈0.05).After therapy of NGF,cell apoptosis in NGF therapy group was higher than that of non-NGF therapy group.Conclusion NGF by periphery can pass cross blood brain barrier and have the protective effect for HIBD in neonatal rats.
