金龙蛇颗粒对原位移植人MKN-45胃癌组织基因表达谱的影响 被引量:5

Effect of Jinlongshe Granule on Gene Expression Profiling in MKN-45 Human Gastric Cancer Orthotopic-transplanted in Nude Mice

摘要 目的:利用基因表达谱芯片研究金龙蛇颗粒对MKN-45胃癌的分子机制。方法:抽提经金龙蛇颗粒治疗的裸鼠原位移植人MKN-45胃癌组织和生理盐水阴性对照的瘤组织总RNA并纯化mRNA,分别逆转录并进行荧光标记,制作成cDNA探针,与包含4096个人类基因的cDNA表达谱芯片进行杂交,筛选出两组间差异表达基因。结果:在mRNA水平上,筛选出包括与DNA结合、转录和转录因子、蛋白翻译、细胞周期等相关表达差异的基因共341条,其中44条(比值>2)表达上调(占12.9%),297条(比值<0.5)表达下调(占87.1%)。结论:金龙蛇颗粒可引起MKN-45胃癌组织一系列基因表达的改变,金龙蛇颗粒抗胃癌的基因途径可能是多环节、多层次的结果。 Objective: To decipher the antitumor mechanisms of Chinese compound Jinlongshe (JLS) granule using cDNA microarray to identify gene expression changes in MKN -45 human gastric cancer. Methods: The total RNAs were isolated from carcinoma tissue of two groups, JLS granule - treated or saline - treated. They were purled, both mRNA were reversely transcribed to the cDNA and labeled with fluorescence to prepare the cDNA probe, which was hybridized with BiostarH -40s human cDNA gene chip to screen the different expressed genes between the two groups. Results: 341 genes among the total 4096 ( 8.33% ) showed expression difference in the cDNA chip assessments. The 297 genes (87.1%, ratio 〈0.5) were down - regulated and 44 gene ( 12.9%, ratio 〉2) were up - regulated, involving multiple scopes as apoptosis, cell cycle, signal transduction, transcription factor, DNA replication, DNA repair, RNA processing and protein synthesis, energy metabolism, immunity - associated, cell receptors as well. Conclusion: The antitumor effect of JLS granule is multi - targeted, which elucidates part of the pharmacologic mechanism of JLS granules in molecular level.
