摘要
目的对15株贵州新城疫病毒进行毒力测定与单克隆抗体分群鉴定。方法测定鸡胚平均死亡时间(MDT)和3日龄脑内接种指数(ICPI),同时对这些毒株的三项表型特征进行了试验观察。并通过间接酶联免疫吸附实验,利用针对NDV的HN基因的单克隆抗体对贵州NDV不同分离株进行分群。结果鸡胚平均死亡时间(MDT)和3日龄脑内接种指数(ICPI)测定表明,FW、F48E8属于速发型毒株;ND98、P1、P2,鸽NDV、Ⅰ系、H2、BY属于中发型毒株,Ⅱ系、L2、P3、ND99、ND79、Lasota属于缓发型毒株。对毒株的三项表型特征试验观察结果表明,血凝解脱速率、血凝素对热的稳定性、ND不同分离株与不同动物红细胞的血凝性等表型特征与NDV毒力没有相关性。间接酶联免疫吸附实验结果表明,A群为速发型强毒株,B群为缓发型弱毒与中等毒力株,C群为介于速发型强毒与中等毒力之间毒株。结论NDV毒力的强弱与血凝素对热的稳定性没有相关性。贵州各地的现场流行毒株,它们的毒力介于强毒与中等毒力之间,与传统的毒力指数测定的结果基本相符。利用三株针对HN蛋白的单抗,由于它们各自的生物学特性和与不同毒力毒株的反应性不同,因而可将用于新城疫病毒强弱毒株的鉴定。
Objective To determine the virulence of different newcastle disease virus (NDV) and classify NDV by monoclonal antibody Methods Mean death time (MDT) and intracerebral pathogenicity index (ICPI) of chick embryo were detected. At the same time, the three phenotypic characteristics of these viruses were determined. Finally, Guizhou' s different NDV strains were divided into three groups by indirect El JSA using monoclonal antibody aiming at HN glycoprotein of NDV. Results The determination of MDT and ICPI showed that FW and F48E8 were classified into velogenic strain. ND98, P1, P2, Bird NDV, Ⅰ strain, H2, and BY belonged to mesogenic strain, while Ⅱ , L2, P3, ND99, ND79, and Lasota were lentogenic strains. The observation of three phenotypic characteristics of these viruses showed that no relativity existed between virulence and the three phenotypic characteristics (hemagglutinin elution, hemagglutinin heat-stability, and different ND strain hemagglutination with different animal's red blood cell). Indirect ELISA indicated that group A was velogenic strain, group B was mesogenic and lentogenic, while group C was prevalent strains whose virulence was between velogenic and mesogenic. Condusion No relativity exists between the virulence of NDV and the hemagglutinin heat-stability. The virulence of the prevalent strain in Guizhou is between velogenic and mesogenic, which is accordant to traditional results. By use of three monoclonal antibodies aiming at HN glycoprotein of NDV, the virulence of NDV can be identified, according to their different biological characteristic and different reactivity.
