摘要 目的:建立敏感的逆转录聚合酶链反应(RT-PCR)方法,研究糖尿病(DM)大鼠视网膜组织中肿瘤坏死因子α(TNFα)mRNA的表达水平。方法:以大鼠两只眼球的视网膜作为1例提取视网膜组织总RNA,用RT-PCR法检测正常大鼠和DM大鼠视网膜组织中TNFα mRNA的含量。结果:正常大鼠视网膜组织中无TNFα mRNA的表达,而DM大鼠视网膜组织中均有TNFα mRNA的表达,且随病程进展TNFα mRNA的表达增加(t=7.413,P<0.05)。结论:RT-PCR法可用于研究大鼠视网膜组织中TNFα mRNA的表达。 Objective:To evaluate the expression of TNFα mRNA by a newly developed sensitive reverse transcriptase polymerase chain reaction(RT-PCR)method in the retina of diabetic rats.Methods:The bilateral eyes of both diabetic and normal rats were enucleated,from which retinal total RNA was extracted.The level of TNFα mRNA was evaluated by RT-PCR in the retinas of diabetic rats and normal control rats.Results:There was no expression of TNFα mRNA in control rats.The expression of TNFα mRNA was found in the retinas of diabetic rats.More expression of TNFα mRNA was detected with progress of the disease(t =7.413,P 〈 0.05).Conclusion:RT-PCR can be used to evaluate the expression of TNFα mRNA in the retina of rats.
