Expression of TGF-β1 and MMP2 in human renal clear cell carcinoma: an immunohistochemical study

摘要 目的研究转化生长因子-β1(TGF-β1)、基质金属蛋白酶2(MMP2)在肾透明细胞癌(RCCC)中的表达及临床意义。方法采用免疫组化SP法测定80例RCCC及30例正常肾组织中TGF-β1和MMP2的表达。80例RCCC中,病理分级:1级19例,2级35例,3级19例,4级7例;临床分期:Ⅰ期35例,Ⅱ期24例,Ⅲ期14例,Ⅳ期7例。结果TGF-β1在RCCC中的阳性表达率为78.8%(63/80),高于正常肾组织的33.3%(10/30);TGF-β1表达与RCCC病理分级、分期呈正相关(P<0.01)。MMP2在RCCC中的阳性表达率为87.5%(70/80),高于正常肾组织的23.3%(7/30);MMP2表达与RCCC病理分级、分期呈正相关(P<0.01)。TGF-β1阳性表达率与MMP2阳性表达率无相关性(P=0.118),但二者的吻合度有统计学意义(κ=0.341,P=0.001);在高分级、高分期RCCC中,二者阳性表达率同时高于低分级、低分期RCCC(P<0.05)。结论TGF-β1、MMP2蛋白的高表达参与了RCCC的发展,可以作为判断RCCC预后的分子指标。 Objective To investigate the expression of transforming growth factor beta 1 (TGF-β1) and matrix metalloproteinases 2 (MMP2) in human renal clear cell carcinoma (RCCC) and their significance. Methods The expression of TGF-β1 and MMP2 in 80 cases of RCCC and 30 cases of normal renal tissue were determined with immunohistochemical SP method. Of the 80 cases of RCCC, 19 were at grade Ⅰ , 35 at grade Ⅱ , 19 at grade Ⅲ, and 7 at grade Ⅳ according to the pathologic grading. Results The expression of TGF-β1 in RCCC was 78.75% (63/80), which was higher than that in normal renal tissue (33.33%, 10/30). The over expression of TGF-β1 was positively correlated with the grade and stage of RCCC( r = 0.672, P 〈 0.01;r = 0.535, P 〈 0.01, respectively); The expression of MMP2 in RCCC was 87.50% (70/80), which was higher than that in normal renal tissue (23.33%, 7/30). The over expression of MMP2 was positively correlated with the grade and stage of RCCC (r = 0.702, P 〈 0.01 ; r = 0.573, P 〈 0.01, respectively). The correlation was found between TGF-β1 and MMP2. The ceexpression of TGF-β1 and MMP2 was higher in high grade and stage than that in low grade and stage (P〈 0.05). Conclusions Up-regulation of TGF-β1 and MMP2 expression may be involved in the progression of RCCC. TGF-β1 and MMP2 can be used as useful molecular markers for prognosis of RCCC.
