The development of bisphosphonates——their chemistry(synthesis,different crystalline hydrates,classification and structure activity relationship),pharmacology(pharmacodynamic properties,mechanism of action,pharmacokinetics and toxicology)and treatment of resorptive bone diseases(including Paget's disease,hypercalcaemia of malignancy,osteolytic bone metastasis and osteoporosis) is reviewed.
