1,6-二磷酸果糖抑制白细胞介素-1β诱导的胰岛β细胞内钙超载

摘要 目的探讨1,6-二磷酸果糖(FDP)对白细胞介素-1β(IL-1β)损伤胰岛β细胞的保护作用。方法分离3-5日龄W istar大鼠胰岛,进行胰岛细胞体外培养,在培养细胞中加入IL-1β及FDP,用紫外分光光度法测细胞内游离钙离子浓度,用火焰原子吸收光谱法测量加入FDP前后细胞的总钙量。结果在胰岛细胞中加入IL-1β后,细胞内游离钙离子浓度及细胞总钙量均升高;在IL-1β损伤后的胰岛细胞中加入FDP的钠盐共同培养后,细胞内游离钙离子浓度及细胞内总钙量均减少(P〈0.01)。结论IL-1β引起胰岛β细胞外钙离子内流入细胞内,导致胰岛β细胞内钙超载。加入FDP可抑制IL-1β引起的胰岛β细胞内钙超载,表明FDP对胰岛β细胞有一定保护作用。 Objective To investigate the protective effect of fructose - 1,6 - diphosphate (FDP) on pancreatic β cells. Methods Pancreas was isolated from three -to- five -day Wistar rats to proceed with cellular culture in vitro. The cytoplasmic free calcium ion concentration was determined by ultraviolet spectrophotometry, the intracellular total calcium by flame atomic absorption spectrophotometry, before and after administration of IL - 1β and FDP. Results The intracellular free calcium ion concentration and intracellular total calcium of pancreatic β cells were increased after the administration of IL - 1β. After the adding of FDP - Na to the pancreatic β cells damaged by IL - 1β, the cytoplasmic free calcium ion concentration and the intracellular total calcium became lower than that in the IL - 1β injury group with-out FDP treatment ( P 〈 0. 01 ). Conclusion Influx of extracellular calcium ions into β cells via voltage - gated calcium channel may induce calcium overload in pancreatic β cells, and FDP can depress this injurious calcium overload, suggesting that FDP could protect β cells from the IL - 1β injury to a certain extent.
