血浆Hcy水平及MTHFR基因C677T多态性与宁夏中青年缺血性脑卒中的关系研究 被引量:1

摘要 目的探讨亚甲基四氢叶酸还原酶(m ethylenetetrahydrofolate reductase,MTHFR)基因C677T多态性、血浆同型半胱氨酸(homocyste ine,Hcy)水平与宁夏中青年缺血性脑卒中的关系。方法应用聚和酶链反应-限制性内切酶片断长度多态性方法(PCR-RFLP)对101例中青年(年龄<55岁)急性缺血性脑卒中患者和96例中青年健康对照者的MTHFR基因C677T多态性进行研究,并测定血浆Hcy水平。结果病例组血浆Hcy浓度为(12.58±8.86)μmol/L,显著高于对照组的(8.96±5.76)μmol/L(P=0.001);两组间基因型分布和等位基因频率无显著性差异(P>0.05)。结论高Hcy血症是缺血性脑卒中的危险因素,未发现MTHFR基因C677T多态性与宁夏中青年缺血性脑卒中的发生存在明显相关性。 Objective To study the relationship between MTHFR gene C677T polymorphism, plasma Hcy level and ischemic stroke in mid - adults and young adults of Ningxia. Methods Polymerase chain reaction and restricted fragment length polymorphism ( PCR - RFLP) method was used to analyze the genotypes of MTHFR gene in 101 patients with ischemic stroke and 96 healthy controls. And plasma Hcy level was determined as well. Results The plasma Hcy level showed significant difference between the patient and control groups (12.58 ± 8.86μmol/L vs. 8.96 ± 5.76 μmol/L, P 〈 0. 01 ) . No significant difference was found in frequencies of genotypes and alleles between two groups ( χ^2 test, P 〉 0. 05 ). Conclusion Elevated plasma Hcy is a risk factor for ischemic stroke, the association between MTHFR gene C677T polymorphism and the incidence of ischemic stroke in Ningxia mid - adults and young adults could not be observed.
