摘要
目的:动态观察脂肪酸代谢及转移酶(SCD-1、CPT-Ⅰ和CD36)在坏死型高脂血症性急性胰腺炎(HLP)大鼠肝脏组织中的表达并探讨其意义。方法:采用胰管内注射5%牛磺胆酸钠诱导高脂血症大鼠产生坏死型HLP大鼠模型。制模后第6、12、24h分别处死大鼠,每组6只大鼠。对照组仅建立坏死型急性胰腺炎大鼠模型。测定血清淀粉酶和胰腺组织病理学评分观察大鼠胰腺组织炎症程度。RT-PCR检测肝脏组织SCD-1、CPT-Ⅰ和CD36mRNA表达;免疫组化检测肝脏组织SCD-1蛋白表达。结果:HLP组大鼠血清淀粉酶水平低于对照组;病理学评分示HLP组大鼠急性胰腺炎程度加重。RT-PCR和免疫组化分析示HLP组大鼠肝脏组织中SCD-1mRNA和蛋白表达显著低于对照组;RT-PCR检测示HLP组大鼠肝脏组织中CPT-Ⅰ和CD36mRNA表达略高于对照组。结论:HLP大鼠肝脏脂肪酸代谢及转移酶异常表达,加重了肝脏组织细胞内外的脂质代谢紊乱,是加重坏死型急性胰腺炎胰腺组织损伤的重要机制之一。
Objective: To investigate the expression of fatty acid metabolic and transferring enzymes in hepatic tissue and to clarify their effects on development of acute necrotizing hyperlipidemic pancreatitis(HI,P) in rats. Methods.-Acute necrotizing HLP was induced in hyperlipidemic Sprague-Dawley rats by intraductal infusion of 5 % sodium taurocholate. The rats with HLP were sacrificed at 6,12,24 hour. The control rats were infused of 5 % sodium taurocholate intraductally. Each group has 6 rats. The serum amylase were determined and severity of pancreatitis was evaluated by histological scoring system. The expression of SCD-1 ,CPT- I and CD36 mRNA in the hepatic tissue were detected by RT-PCR,the expression of SCD-1 protein was detected by immunohistochemical staining. Results:As compared with the controls,the serum amylase level was decreased and the severity of pancreatitis was worsen in the HLP rats. In contrast to the controls, the hepatic expression of SCD-1 mRNA was down- regulated and the SCD-1 protein was negative in HLP rats; the hepatic CPT- I and CD36 mRNA was up-regulated in HLP rats. Conclusions:The abnormally expressing of the hepatic fatty acid metabolic and transferring enzymes was involved in aggravating the pancreatic lipids metabolic disturbance and worsening the acute necrotizing HLP.
