铜绿假单胞菌注射液对K562细胞株处理后淋巴细胞凋亡及免疫功能的影响 被引量:8

摘要 目的观察铜绿假单胞菌注射液对肿瘤微环境中正常人外周血淋巴细胞的凋亡的抑制作用及免疫增强作用。方法分离纯化正常人外周血淋巴细胞并与K562细胞株上清共培养,与不同浓度的铜绿假单胞菌注射液作用72h后,用酶标仪或流式细胞仪进行各项指标检测,观察铜绿假单胞菌注射液对人外周单个核细胞(PMBC)增殖率、凋亡率、坏死率、凋亡相关蛋白及细胞表面蛋白表达的影响。结果铜绿假单胞菌注射液能增强K562细胞株上清作用后淋巴细胞增殖,降低细胞凋亡率、坏死率,降低促凋亡蛋白Fas的表达,增加抑凋亡蛋白Bcl-2的表达,增加CD25、CD69的表达,对CD4+/CD8+无显著影响。结论铜绿假单胞菌注射液可以抑制肿瘤微环境中正常人外周血淋巴细胞的凋亡、坏死,并能增强淋巴细胞的免疫功能。  OBJECTIVE To investigate the inhibitory effects of Pseudomonas aeruginosa injection on the lymphocytes apoptosis and the immune enhancing effects on peripheral lymphocytes of volunteers.METHODS The lymphocytes were isolated from the volunteers,and these lymphocytes were co-cultured with the supernatants of K562 cell lines. After being incubated with different concentrations of Pseudomonas aeruginosa injection for 72 h, the proliferation of lymphocytes was detected by microplate reader, and the apoptosis, necrosis, expression of Fas, Bcl-2, CD25 and CD69, the CD4 positive cells and CD8 positive cells were analyzed by flow cytometry.RESULTS Pseudomonas aeruginosa injection increased the proliferation of the lymphocytes co-cultured with the supernatants of K562 cell lines and the expression of Bcl-2, CD25 and CD69. The apoptosis and necrosis in lymphocytes, the expression of Fas were reduced by Pseudomonas aeruginosa injection significantly. But Pseudomonas aeruginosa injection affected little on the ratio of CD4+ lymphocytes to CD8+ lymphocytes.CONCLUSION Pseudomonas aeruginosa injection can inhibit the apoptosis and necrosis in lymphocytes co-cultured with the supernatants of K562 cell lines, and can enhance the immune function.
