摘要
采用紫外分光光度法、TBA法、琼脂糖凝胶电泳法检测维生素C或H2O2体系对DNA的作用,发现DNA在260nm处吸光值下降,检测到DNA的脱氧核糖损伤,观察到DNA链降解.痕量的过渡金属离子Fe2+等能加速上述反应的进行,各种HO·清除剂可抑制上述反应.表明在上述两类反应体系中产生了HO·,是HO·破坏DNA的碱基和脱氧核糖。
DNA damage by oxygen radical was studied in the systems containing ascorbate or H 2O 2, using ultraviolet-spectrometer, TBA reaction and agarose gel electrophoresis. The absorption in 260nm of DNA decreased. Deoxyribose damage was found in TBA reaction. DNA backbone cleavage was observed in agrose gel electrophoresis. Trace metal ion such as Fe 2+ and so on could accelerate the reactions mentioned above. In all case DNA damage was prevented by a series of HO · scarvages. The role of HO · induced by the above two systems in the DNA damage including base, deoxyribose and backbone degradation was discussed.
