摘要
目的:建立以反相高效液相色谱法测定肾移植患者全血中环孢素A(CsA)浓度的方法。方法:采用单步萃取法提取,以反相高效液相色谱法测定,其中色谱柱为C8,流动相为乙腈-甲醇-水(45∶30∶25),流速为1.2mL·min-1,检测波长为210nm,柱温为60℃。结果:全血中CsA的检测浓度在50~600ng.mL-1范围内线性关系良好(r=0.9997);方法平均回收率为99.44%,日内、日间RSD均<4%;当S/N≥3时,方法的最低检测限为10ng.mL-1。结论:本方法准确、专一、回收率高、受代谢物干扰少、相对成本较低,可用于临床血药浓度检测及药动学研究。
OBJECTIVE: To establish a RP - HPLC method for the determination of the concentration of cyclosporin A (CsA) in whole blood of patients undergoing renal transplantation .METHODS: CsA was extracted from whole blood by single- step extraction and determined by RP - HPLC, and separated on a C8 column with a mobile phase consisted of acetonitrile- methanol- water(45 : 30 : 25) at a flow rate of 1.2mL·mL^-1.The detective wavelength was 210nm and the column temperature was maintained at 60℃ .RESULTS: The linear range of CsA was 50-600ng·mL^-1( r = 0.999 7) .The average recovery rate of CsA was 99.44%, with both intra - day and inter - day RSD less than 4% .When S/N≥3, the detection limit of CsA was 10ng ·mL^-1.CONCLUSION:The established method was accurate and specific, meanwhile,it has high recovery,little interference and low cost, thus applicable for the determination of blood concentration and pharmacokinetics of CsA.
