摘要
目的:肾精亏虚对小鼠视网膜细胞凋亡相关基因Bcl-2、Bax的影响。方法:将30只雄性昆明小鼠用计算机取随机数法随机分为正常对照组、模型组和补肝组3组。其中模型组和补肝组采用惊恐加房劳的复合伤肾法制造肾精亏虚模型。补肝组每天给予0.3mL/10g体重的一贯煎浓缩液灌胃。正常对照组及模型组予等量生理盐水灌胃,连续给药21天。制备小鼠视网膜石蜡切片行免疫组化ABC法染色,并对染色结果进行计算机图像分析系统观察分析。结果:Bcl-2和Bax在正常组阳性反应少见,且只出现于节细胞血管;模型组小鼠视网膜神经节细胞层(GCL)和内核层(INL)可见大量的阳性细胞;补肝组小鼠视网膜神经节细胞层(GCL)和内核层(INL)可见少数阳性反应细胞。各组小鼠视网膜细胞Bcl-2和Bax表达的阳性细胞率比较:模型组与正常组及补肝组比较有非常显著的差异(P<0.01);正常组与补肝组比较无显著的差异(P>0.05)。结论:凋亡相关基因Bal-2和Bax在"惊恐、房劳"因素复制的肾精亏虚模型小鼠视网膜的表达增强,二者在肾精亏虚模型小鼠视网膜细胞凋亡中起重要作用。
Objective： To research the expression of apoptosis -related genes Bcl -2 and Bax in the retinas of mice which has insufficient ShenJing The liver and kidney promote and reinforce each other,deficiency of the one will lead to deficiency of the other,and the physiologic and pathologic condition of the liver may be reflected in the changes of the eyes. After the hypothesis, we researched the affect of mice which has insufficient ShenJing that will lead to deficiency of the liver and the mechanism of these affects, the models were made by the ＂affair combined with excessive sexual behavior＂. Methods： The mice were allocated to 3 groups（ blank control group, model group,Bugan group） by random number. The mice of the model group and the Bugan group were processed by the ＂affair combined with excessive sexual behavior＂ to made the model of. insufficient ShenJing. The mice in Bugan group were lavaged of concentrated solution of Yi Guan prescription, the dosage is 0.3mL/10g based on their weigh. Other groups treat with physiological saline at the same dosage. This phase would continued 21 days. To reseaech the expression of aopotosis - related genes（ Bcl - 2, Bax） in retinas. Result： The express of Bc1-2 and Bax in the GCL and INL of retina is geting increased, the positive reaction of model mice increased significant compared with cotrol and Bugan group（ P 〈 0.01 ）. Conclusion： The deficiency of the kidney will lead to deficiency of the liver, may damage the retinal , maybe it influence the ratio of aopotosis - related genes Bel -2 and Bax, and the apoptosis of retina is rised.
