阿魏酸钠对肺纤维化大鼠肺组织Smad4的影响及意义 被引量:1

摘要 目的观察阿魏酸钠对肺纤维化大鼠肺组织Smad4 mRNA表达的影响,探讨其治疗肺纤维化的作用及机制。方法将30只SD大鼠随机分为对照组、肺纤维化组和阿魏酸钠治疗组,每组10只。肺纤维化组和阿魏酸钠治疗组以气管内滴入博来霉素(5mg/kg)建立肺纤维化动物模型,治疗组采用阿魏酸钠(150mg/kg体重)灌胃治疗。苏木精-伊红染色检测肺组织病理改变;免疫组化检测Ⅰ型胶原纤维;原位杂交检测Smad4 mRNA的表达。结果与对照组比较,肺纤维化组Ⅰ型胶原纤维(0.97±0.34比0.27±0.08,P<0.001)和Smad4 mRNA(0.17±0.10对0.07±0.03,P<0.01)的表达均显著增高。阿魏酸钠治疗组与肺纤维化组比较,Ⅰ型胶原纤维(0.39±0.25比0.97±0.34,P<0.001)和Smad4mRNA(0.11±0.03对0.17±0.10,P<0.05)的表达均显著降低。结论阿魏酸钠可能通过抑制大鼠肺组织Smad4 mRNA的表达,从而具有抗肺纤维化的作用。 Objective To investigate the effects of sedium ferulate on the expression of Smad4 mRNA in lungs of rats with pulmonary fibrosis,and explore the mechanism of sodium ferulate on pulmonary fibrosis.Methods Thirty SD rats were randomly divided into control group, pulmonary fibrosis group and sodium ferulate treatment group. A rat model of pulmonary fibrosis was established by the iraratrachead injection of bleomycin(5mg/kg). Sodium ferulate were administered intragastricly in the treatment group with a dose of 150mg/kg.The lung histopathology was examined by HE staining,type Ⅰ collagen quantity was examined by immunohistochemical assay and Smad4 mRNA was tested by method of in situ hybfidzation(ISH) .Results Compared with the control group,the expression of type Ⅰ collagen quantity and Smad4 mRNA in the pulmonary fibrosis group were significantly higher(0.97 ± 0.34 vs 0.27 ± 0.08, P 〈 0.001 ;0.17 ± 0.10 vs 0.07 ± 0.03, P 〈 0.01, respectively ). While compared with the pulmonary fibrosis group, the expression of type I collagen quantity and Smad4 mRNA in sodium femlate treatment group were significantly lower(0.39 ± 0.25 vs 0.97 ± 0.34, P 〈 0.1301 ;0.11 ± 0.03 vs 0.17 ± 0.10, P 〈 0.05, respectively ). Conclusion Sodium femlate may has a treatment effect on pulmonary fibrosis through inhibiting the expression of Smad4 mRNA in the lungs of rats with pulmonary fibrosis.
