摘要
目的探讨去甲肾上腺素转运体(NET)基因T-182C和G1287A多态性与中国汉族人群重性抑郁症(MD)之间的关系。方法采用性别、年龄1∶1配对的病例对照设计,纳入符合美国精神障碍诊断与统计手册第4版(DSM-Ⅳ)MD诊断标准的患者152例和正常对照152名。应用聚合酶链反应(PCR)和DNA直接测序技术,检测NET基因的T-182C和G1287A多态性,并进行关联及单倍型分析。结果①患者组与对照组间T-182C多态性基因型和等位基因频率分布差异无显著性(Х2=3.934,P>0.05;Х2=1.738,P>0.05),两组间G1287A多态性基因型和等位基因频率分布差异亦无显著性(Х2=2.545,P>0.05;Х2=1.502,P>0.05);②单倍型分析显示两组间上述两个位点的C-A单倍型频率具有显著性差异(Х2=4.045,P<0.05,OR=1.889),患者组C-A单倍型频率明显高于对照组。结论去甲肾上腺素转运体基因可能是中国汉族人群重性抑郁症的易感基因。
Objective To investigate the association between the two genetic polymorphisms of T-182C and G1287A of norepinephrine transporter gene(NET) and major depression(MD) in Chinese Han population. Methods One hundred and fifty two cases were diagnosed by DSM-IV, 152 controls were well matched to cases group on gender, age and ethnicity, who had no past or present history of major psychiatric disorders. Polymerase chain reaction (PCR) and DNA direct sequen- cing technique were used to detect the T-182C and G1287A genetic polymorphism of the NET gene. The frequency distribution of genotypes and alleles in the two groups was analyzed, and the association and haplotype analysis was made simultaneously. Results (1)There were no significant differences in the genotype transporter distributions or allele frequencies of the T-182C genetic polymorphism (X^2 =3. 934, P 〉0. 05 ;X^2 = 1. 738, P 〉 0. 05 ) , and no significant differences in the genotype distribution or allele frequencies of the G1287A genetic polymorphism between cases and controls ( X^2 = 2. 545, P 〉 0. 05 ; X^2 = 1. 502, P 〉0. 05). (2)A weak association between C-A haplotype and major depression was found ( X^2 = 4. 045, P = 0. 044, OR = 1. 899), the C-A haplotype frequency in cases was higher than that in controls. Conclusions Norepinephrine transporter gene is one possible suspected gene for major depression in Chinese Han population.
