摘要
[目的]研究2型糖尿病膀胱逼尿肌收缩功能的改变和M3受体表达的变化。[方法]建立2型糖尿病大鼠模型,在糖尿病病程第16周,应用离体膀胱灌注方法观察逼尿肌收缩功能的变化;RT-PCR和Western blotting方法观察逼尿肌M3受体mRNA表达的变化。[结果]2型糖尿病组逼尿肌收缩功能低于正常对照组,2型糖尿病两组大鼠膀胱内压为(17.61±3.62)cmH2O/100mg,正常对照组为(24.57±4.20)cmH2O/100mg;2型糖尿病组逼尿肌M3受体mRNA和蛋白的表达均高于正常对照组,分别为63.53%±3.54%和39.37%±3.70%,43.53%±1.59%和25.39%±1.40%。[结论]2型糖尿病大鼠逼尿肌M3受体的生物合成是上调的,但是逼尿肌的收缩功能却降低,可能是M3受体功能方面存在着一定的障碍。
Objective: To investigate on the changes of contractile function of detrusor muscles of type 2 diabetes mellitus (DM) rats and expression changes of M3 receptor of detrusor muscles. Methods:Type 2 DM model rats were prepared. Changes of contractile function of detrusor muscles were observed by using isolated irrigation of bladder at the 16th week of DM course. Changes of m - RNA and M3 receptor proteins of rats were observed by adopting RT-PCR and Western blotting method. Results: The contractile function of detrusor muscles in type 2 DM group was worse than that in control group, internal pressure of bladder in type 2 DM group was (17.61 ± 3.62 ) cm H20/100 mg, and (24.57 ± 4.20) cmH2O/100mg in normal control group; expression of m - RNA and M3 receptor protein of detrusor muscles in type 2 DM group was higher than that in normal control group, respectively(63.53 ± 3.54)% and (39.37±3.70)% and (43.53±1.59)% and(25.39± 1.40)%. Conclusion: Biosynthesis of M3 receptors of detrusor muscles of type 2 DM rat is up - regulated. But the contractile function of detrusor muscles decreased. The reasons are supposed possibly that there were some disorders existed in M3 receptor function.
