白细胞介素-6和肿瘤坏死因子α对大鼠下丘脑CRH神经元兴奋作用的研究 被引量:8

摘要 为证明下丘脑产生的促肾上腺皮质激素释放激素(CRH)与白细胞介素-6(IL-6)和肿瘤坏死因子α(TNFα)的中枢作用有关,应用免疫组织化学方法,通过观察即早基因c-fos的表达,对IL-6和TNFα经脑室引入后对下丘脑CRH神经元的兴奋作用进行了研究。结果显示,在脑室注射IL-6或TNFα5h后,下丘脑室旁核小细胞部出现强烈Fos表达。免疫组织化学双标记法显示,两种细胞因子诱导的Fos表达细胞中均有许多细胞同时呈现CRH免疫反应。相比之下,双标细胞数量在IL-6引入者较TNFα引入者多。该结果表明。 In order to demonstrate the involvement of corticotropin releasing hormone (CRH) in the central effect of IL 6 and TNFα. The present study investigated immunohistochemically the Fos expression in hypothalamic CRH containing neurons following intracerebroventricular (Icv) administration of IL 6 and TNFα. A strong Fos expression was found in the parvocellular subdivision of paraventricular nucleus (PVN) 5 hours after IL 6 or TNFα injection. Fos and CRH double immunohistochemical staining revealed that a part of Fos expression neurons induced both by IL 6(Icv) or TNFα (Icv), belonged to CRH containing neuorns. More double labeled cells were found in the case of IL 6 injection. The present result indicated that the hypothalamic CRH containing neurons could be activated in different degree by centrally administration of IL 6 and TNFα.
