摘要
目的:初步观察人口腔白斑及正常黏膜组织中蛋白质的差异表达,为进一步探讨口腔白斑发病的分子机制奠定基础。方法:应用蛋白质组学的双向电泳技术,对口腔白斑和正常黏膜组织中蛋白质表达进行比较,观察其差异点。结果:得到了分辨率和重复性均好的人口腔白斑及正常黏膜组织的双向凝胶电泳图谱,平均蛋白质点数分别为(1726±67)个和(1608±73)个。发现人口腔白斑与正常黏膜组织蛋白表达有明显差异,差异表达蛋白质点数共38个,主要是蛋白表达量的增加和减少,其中16个点在白斑中为高标达,22个点在白斑中为低表达。结论:口腔白斑发生发展可能与口腔黏膜组织中某些蛋白质的表达和修饰改变有关,对这些差异蛋白质点的进一步分析有助于研究口腔白斑发病机制及分子标记。
Objective: To identify differential proteins expression of human oral leukoplakia and paired normal oral mucosa. Methods : The differential proteins expression of oral leukoplakia and paired normal oral mucosa tissues were characterized by means of two - dimensional gel electrophoresis (2 - DE). Results : The well - resolved, reproducible 2 - DE patterns of oral leukoplakia and normal oral mucosa tissues were obtained. And average spots were 1726 ± 67, 1608 ± 73 in oral leukoplakia and normal oral mucosa tissues respectively. The 38 differential protein spots were identified. There were 16 protein spots in oral leukoplakia were higher expressed than those of oral mucosa. 22 protein spots in oral leukoplakia were lower expressed than those in normal oral mucosa. Conclusion: The development of oral leukoplakia might due to the expression and modification of some proteins in oral mucosa tissues. The study on these differential proteins expression will help to screen special biomarkers of oral leukoplakia.
