摘要
利用微波诱变技术对产α-乙酰乳酸脱羧酶的枯草芽孢杆菌W195进行了微波诱变效应的研究。通过对微波辐照不同时间及不同强度的实验,对突变菌株的α-ALDC产量进行比较和分析,得出诱变剂量和诱变效应的关系及诱变的最佳剂量,同时测定所得较高产量突变菌株的遗传稳定性。研究结果表明,诱变剂量越大,菌体的致死效应越大,而诱变效应是先增大后减小,存在一个最佳值。本实验最佳诱变剂量为微波低强度间歇辐照40s,得到的突变株M3-14产量为0.375,相对出发菌株提高了55%,遗传性状基本稳定。
To examine the mutagenic effect of microwave radiation, Bacillus subtilis W195 was treated by microwave. In experiments W195 was treated by different time and different intensity of microwave radiation, Based on the comparison and analysis of the yield of α- ALDC of the mutants, the optimal dosage was obtained, and the genetic stability of the mutants was determined. The results showed that the optimum dosage of irradiation was small fire of 40 seconds, and M3-14 comparatively yield (OD522)of α-ALDC was higher than 0.375,which had been increased about 55% than that of the initial strain.
