摘要
目的探讨17-β-雌二醇(E2)在体外对人B淋巴细胞系IM-9细胞的调节作用。方法四甲基偶氮唑兰(MTT)法检测不同浓度E2对IM-9细胞增殖时间和剂量依赖性的影响;逆转录多聚酶链反应(RT-PCR)检测免疫球蛋白G(IgG)的分泌。结果48 h后低浓度E2(≤10-7mol.L-1)能明显地促进IM-9细胞的增殖(P<0.05),并呈时间-剂量依赖性,而高浓度E2(>10-7mol.L-1)对IM-9细胞有明显的抑制作用(P<0.05),也呈时间-剂量依赖性;RT-PCR扩增IgG显示低浓度的E2(10-10mol.L-1)明显促进IgG分泌(P<0.05),高浓度的E2(10-6mol.L-1,10-4mol.L-1)明显抑制IgG的分泌(P<0.05)。结论E2能调节IM-9淋巴细胞增殖,同时影响免疫球蛋白的分泌。
Objective To investigate the modulation of 17-β-estradiol( E2 ) on human B lymphocyte series IM-9 cells in vitro. Methods After stimulated from the different concentration of E2 , the IM-9 cells proliferation were detected by methyl thiazolyl tetrazolium(MTT) and the cDNA of IgG were examined by reverse transcription-polymerase chain reaction (RT-PCR) to explain the influence of E2 on the secretion of immunoglobulin. Results Low concentration of E2 (≤ 10^-7 mol · L^-l ) could obviously promote the proliferation of IM-9 cells in time-dose-dependent after 48h. However( P 〈 0.05 ), higher concentration ( 〉 10^-7mol·L^-1) of E2 inhibited the proliferation of IM-9 cells obviously(P 〈 0. 05),which also was time-dose-dependent. The fragment of IgG cDNA detected by RT-PCR showed that low concentration( 10^-10mol·L^-1) could obviously promote the secretion of IgG ( P 〈 0.05 ), but the group of higher concentration ( 10^-6 mol·L^-1, 10^-4 mol·L^-1) suppressed the secretion of IgG (P 〈 0.05). Conclusion E2 can modulates the IM-9 cell proliferation and affect the secretion of immunoglobulin.
