摘要
通过对影响长豇豆AFLP分析技术体系的若干关键因素,如DNA的提取方法、酶切与连接时间、预扩增和选择性扩增以及电泳和染色条件等的研究,建立一套经优化的长豇豆AFLP分析技术体系。应用该体系进行长豇豆AFLP分析,能够得到清晰的分子标记图谱,且结果稳定,重复性好。
Several main factors affecting the results of AFLP fingerprints for asparagus bean （Vigna unguiculata ssp. sesquipedlis （L.）Verd. ） were studied, which including the method of genomic DNA extraction, enzyme digestion and ligation, pre-amplification and selective-amplification, electrophoresis, dyeing conditions etc. An optimized AFLP analysis system was established. Using this system, a clear, credible and repeatable AFLP fingerprints could be obtained.
