建立高效液相指纹图谱分析无机离子对丹参不定根有效成分影响 被引量:5

The Fingerprint for Adventitious Root of Radix Salviae Miltiorrhizae Developed by High-performance Liquid Chromatography and its Active Component Primary Analysis

摘要 目的:建立丹参不定根RP-HPLC指纹图谱分析方法,分析无机离子对丹参不定根影响。方法:高效液相色谱法,RP-Waters C18(150 mm×3.9 mm,5μm),流动相为0.5%醋酸甲醇和0.5醋酸%水梯度洗脱,流速1.0 mL.min-1,检测波长:270nm,柱温:30℃。对丹参不定根进行指纹图谱分析。结果:在此色谱条件下,3组共9份样品的不定根可检测出11个相对位置稳定的共有峰,其中确定9个共有峰作为成分变化指示峰,并确定了5个已知成分,结果显示无机金属离子能明显提高不定根中丹酚酸类成分的含量,而对丹参酮等脂溶性成分影响不显著。结论:该方法稳定可靠,重复性强,可用于评价对丹参不定根次生代谢成分变化情况,为丹参不定根的培养及进一步研究奠定基础。 Objective:To establish RP-HPLC fingerprint for adventitious root of Radix Salviae Miltiorrhizae and primarily investigate effects of different mental ion on it. Method. The Methanol extraction of Radix Salviae Miltiorrhizae was filtered through 0.45 mm filters and separated by using a Resolve TM C18 column (Waters, 150mm × 3.9 mm, 5 μm) at 30℃. The mobile phase consisted of acetonitrile and water was used as gradient elution for the separation. The flow rate was set at 1.0 mL·min^-1 and detection was set at 270 rim. The fingerprint was then established as above condition for adventitious root of Radix Salviae Miltiorrhizae Result: 11 peaks in common of 9 samples of treated with 3 kind of metal ion to adventitious root were acquired and 9 of them were chosen as symbolization for the component of Radix Salviae Miltiorrhizae. Conclution:The fingerprint profile provides a good approach to value the quality and monitor the change of content of active component in adventitious root of Radix Salviae Miltiorrhizae.
