摘要
目的:采用小鼠异位成骨技术及甲基麝香草酚蓝比色法检测重组人骨形态发生蛋白-2(rhBMP-2)的生物学活性。方法:将rhBMP-2埋入小鼠肌间隙内,14d后取出新生组织,采用血清钙试剂盒检测其钙含量。结果:随着给药组剂量递增,相应地钙含量也增加,二者具有较强的量效关系。结论:此方法为本实验室独创,较传统的血清碱性磷酸酶方法更为方便、快捷,是一种能够定量检测rhBMP-2活性的新方法。
Objective: Technique of allotopic osteogenesis in mouse and serum calcium diagnostic kit(methyl-thymol blue colormetric method) were used to determine the biological activity of recombinant human bone morphogenic protein-2 (rhBMP-2). Methods: rhBMP-2 in powder form was embedded in intermuscular spatium of posterior limb. 14 days after being embeded, neoplasm in operation sites was removed and ground, and calcium content in neoplasm was measured with serum calcium diagnostic kit. Results: It showed that the calcium level increased in parallel to that of implanted rhBMP-2 in dose-dependent manner. Conclusion: A new method for rhBMP-2 activity determination is established, which is more feasible and sensitive than traditional alkaline phospatase(AKP) method.
