摘要
Sry基因是哺乳动物性别决定的开关基因,其功能是调控下游Dmrt、Sox3、Sox9等基因来启动性别的分化过程。Dmrt家族是一个在性别决定和分化发育中具有重要功能的基因家族,该家族成员都含有一个具有DNA结合能力的保守基序-DM结构域。本文通过PCR及克隆获得了江豚的Sry基因全序列;通过PCR-SSCP法筛选获得了江豚的Dmrt家族六个不同DM序列。结果显示,雄性个体中存在Sry基因,而Dmrt基因在雌雄个体中都有,与其他动物相关基因进行聚类分析,显示它们在进化上具有高度的保守性。
The Sty( sex determining region on Y chromosome) gene is master switch in sex determining of mammal. The function is start sex determination and differentiation up through work upon Dmrt,Sox3 ,Sox9 and so on. The Dmrt genes constitute a new gene family related to sex-determination. They encoded transcription factors characterized by a conserved zinc finger-like DNA-binding motif, the DM domain. In this study, the PCR technique was used to amplify the Sry complete sequence and the conserved DM domains of Dmrt genes in Neophocaena phocaenoides, then, a sequence (760 bp ) was identified and named as Npsry in male, six different DM sequences ( 140 bp)were identified and named as NpDmrtl , NpDmrt l a, NpDmrt3 , NpDmrt3α, NpDmrt4. and NpDmrt5 according to its name Neophocaena phocaenoides. The results indicated that Sry gene and Dmrt gene were all highly conservative in phylogeny, and the Dmrt genes have several members.
