摘要
[目的]研制李斯特菌显色培养基。[方法]合成环己酮七叶苷-β-D-葡萄糖苷(CHE-β-D-葡萄糖苷)。通过22种菌对其检测效果进行验证。[结果]环己酮七叶苷-β-D-葡萄糖苷与七叶苷具有相同的检出效能;但其显色效果优于七叶苷,在环己酮七叶苷-β-D-葡萄糖苷琼脂培养基上,被分解的游离环己酮七叶苷不扩散,固定在细菌菌落上,形成黑色菌落,从而有利于准确地鉴别和计数。与此相反,七叶苷琼脂上的阳性菌落,其分解产生的黑色素向周围培养基扩散,难以分辨阳性和阴性菌落,影响鉴别和计数的准确性。[结论]与传统七叶苷显色培养基相比,环己酮七叶苷-β-D-葡萄糖苷显色培养基对分离和鉴定细菌存在很大优越性。
This is to develop a chromogenic media for isolation and identification of listeria. The chromogemc media of cyclohexenoesculetin-β-D-glucoside was produced; its efficiency was evaluated by 22 reference bacteria. The efficiency of cyclohexenoesculetin-β-D-glucoside was equal to that of esculetin, but its effect was superior to that of esculetin, because the black chromogenic substrate on cyclohexenoesculetin-β-D-glucoside agar didn't diffuse and positive bacteria formed distinct colonies that could easily be identified easily. On the contrary, the black substrate on esculetin agar diffused easily and positive colonies couldn't be identified appropriately. Compared with traditional chromogenic media of esculetin, the chromogenic media of cyclohexenoesculetin-β-D-glucoside has advantage of isolation and identification.
