摘要
以香榧(Torreya grandis)的叶片和胚乳为材料,利用改进的CTAB-硅珠吸附法提取DNA,分别就叶片与胚乳建立了香榧的AFLP银染反应体系。结果表明:采用EcoRⅠ/MseⅠ酶切体系,64对引物中有36对引物既不适合叶片也不适合胚乳;适合胚乳AFLP分析的引物有20对,适合胚乳叶片AFLP分析的引物组合有15对,既适合胚乳又适合叶片的引物有7对。实验结果为利用AFLP标记对香榧进行深入研究打下了基础。
Using DNA extracted by CTAB-silicon-based method from leaves and endosperm of Torreya grandis, a silver staining-based AFLP system was established for leaves and endosperm respectively. The result showed that using the digestion system of EcoRⅠ/MseⅠ, 36 of 64 primer pairs were not suitable for both leaves and endosperm, 20 primer pairs suitable for endosperm, 15 primer pairs suitable for leaves, and 7 primer pairs suitable for the both. This results provides fundamentals for further molecular studies of Torreya grandis using AFLP marker.
