摘要
目的研究格列美脲对健康新西兰白兔心肌缺血预适应的影响。方法以健康新西兰白兔为实验对象,通过开胸结扎左冠状动脉前降支的方法建立心肌缺血预适应动物模型。将实验动物随机分为对照组、预适应组、格列美脲组和格列美脲+预适应组4个组别。实验过程中记录心电图变化,实验结束后,测量血肌钙蛋白,取心脏大体TTC染色,计算心肌梗死面积,以肌钙蛋白和心肌梗死面积作为终点效应指标,评价药物对心肌缺血预适应的影响。结果预适应组与对照组相比,实验后血肌钙蛋白、心肌梗死面积较前者明显降低,单用格列美脲不影响血肌钙蛋白和心肌梗死面积,而格列美脲+预适应组与预适应组相比,实验后血肌钙蛋白、心肌梗死面积无明显差异。结论格列美脲对新西兰白兔心肌缺血预适应没有影响。
Objective To investigate the effect of glimepiride on myocardial ischemic preconditioning in healthy Newzealand white rabbits. Methods The heathy Newzealand white rabbits were divided into four groups randomly. A thoracotomy was performed to copy IPC and ischemic/reperfusion models. Tighten left anterior descendening branch of coronary artery to produce regional ischemia and then release allowing reperfusion. During the experiments, ECG of limb leads were recorded. After experiments, the sizes of myocardial infarction area were calculated and the concentration of troponin T was detected. Results The sizes of myocardial infarction area and troponin T level were lower in IPC group than that of control group (P 〈 0.05). There was no difference in glimepiride group and control group. Similarly, there was no difference in glimepiride + IPC group and IPC group. Conclusion Glimepiride has no adverse effects on IPC in the healthy Newzealand white rabbits.
