摘要 目的:探讨安迪(Adi)注射剂对人白血病K562细胞株增殖和凋亡的影响.方法:MTT法测定K562在Adi作用后的细胞活力(A值);流式细胞技术检测细胞凋亡变化;电镜观察细胞的形态学改变等;综合评价Adi对K562细胞株增殖和凋亡的影响.结果:①MTT法测得Adi在(2.5~20.0)μg/mL浓度范围内能明显抑制K562细胞的增殖,并具有浓度依赖性,20.0μg/mL Adi作用36h的K562细胞株A值最低;②流式细胞仪检测结果显示10.0,20.0μg/mLAdi处理组K562细胞的凋亡率分别为50%和39%,明显高于对照组的10%(P〈0.05);③电镜下可见K562细胞呈典型凋亡样改变.结论:Adi能明显抑制K562细胞增殖,并具有显著促进细胞凋亡的作用. AIM: To observe the effect of Andi injection (Adi) on proliferation and apoptosis of erythroleukemia K562 cells. METHODS: The effect of Adi on proliferation and apoptosis of K562 cells was assayed by using MTT assay to measure the cell vitality ( A value ) , flow cytometer to detect cell apoptosis, electron microscopy to observe the morphological changes after the action of Adi. RESULTS: ① MTT method showed that within the range of (2.5 - 20.0) μg/mL, Adi obviously inhibited the proliferation of K562 cells in a dose-dependent manner, and 20.0μg/mL Adi which affected K562 cells for 36 h had the lowest A value; ② The apoptosis rates were 50% and 39% in the cells that had been treated with 10.0,20.0 μg/mL Adi, respectively. The apoptosis rates were significantly higher than that of control group ( 10%, P 〈 0. 05 ) ; ③ Typical apoptotic changes were found in K562 cells treated with Adi by electron microscope. CONCLUSION: Adi can inhibit the leukemic cell proliferation and induce the apoptosis of K562 cells.
