摘要
为了提高犬的繁殖力,建立完善的犬人工授精体系和保存优秀的犬精子,本研究对8只犬进行了采精和精液的超低温冷冻保存试验,并对犬的精液超低温冷冻保存液进行了系统的筛选。结果表明,渗透压450mOsm/L的配方V(葡萄糖1.98g、蔗糖3.42g、柠檬酸钠0.59g、谷氨酸钠0.75g、三羟甲基氨基甲烷1.21g、维生素B_610mL、维生素B_(12)10mL、卵黄20mL和甘油8mL)对犬精液冷冻保存的效果最佳,解冻后精子的活率为0.58,畸形率为17.2%,满足输精和进行超低温冷冻保存精子的要求。
In order to increasing the reproductive capacity of dog, perfecting artificial insemination( AI) technology and preserving spermatozoa of excellent dog. We had collect and frozen the semen of eight dog, at the same time, had a systemic filtrate to the spermatozoa freezing--extender of dog. The result showed that the extenderV (composing by glucose 1.98g, sucrose 3.42g, citric acid sodium 0. 59g, glutamic acid sodium 0.75g, EDTA--Na2 1.21g, vitamin 10ml, vitamin Blz 10ml,yelk 20ml and glycerol 8ml) is the best cryopreservation extender of dog. The forward motility sperm is 0. 58, pathomorphology sperm is 17.2%, meeting the quality criteria apply for AI and storage at-196℃.
