摘要
以一氯苯为唯一碳源,从某化工厂废水处理厂的活性污泥中分离得一株一氯苯降解菌,初步鉴定属于气单胞菌属(Aeromonas)。用气相色谱法测得该菌在3周内的氯苯降解率为33%。不同的质粒消除试验结果表明,该菌质粒很稳定。已成功地将质粒转移到大肠杆菌(E.coliJF803)中。该转化子能利用一氯苯为唯一碳源生长,且转化子中有与原菌株电泳迁移率相同的质粒。因此,可以认为该质粒携带有降解一氯苯的基因。
A chlorobenzene degradative strain was isolated from activated sludge by using chlorobenzene as sole carbon source. It was preliminarily identified as an Aeromonas . The degradation rate of chlorobenzene was 33% in 3 weeks tested with gas chromatography. Different methods were used to cure the plasmids and it was found that the plasmids were very stable. The plasmid was transferred to E. coli. The transformant could use chlorobenzene as sole carbon source and plasmid was detected with the same electrophoretic transference rate as the original strain. Thus, it can be confirmed that this plasmid possesses the necessary gene for chlorobenzene degradation.
