七叶皂苷对胰腺炎大鼠脑损害保护作用的实验研究 被引量:3

摘要 目的研究七叶皂苷(aescine)对重症急性胰腺炎模型大鼠脑组织损害的保护作用。方法72只雄性Wistar大鼠随机分为三组,每组24只:假手术组(对照组),重症急性胰腺炎组(模型组),七叶皂苷干预组(干预组)。采用胰管穿刺注射5%牛磺胆酸钠制备大鼠重症急性胰腺炎模型。分别在术后第3,6和12小时采取脑组织标本,采用免疫组化染色法检测脑组织核因子-κB(nuclear factor-κB,NF-κB)活性,同时应用放射免疫法检测脑组织肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)蛋白水平,并测定脑组织含水量及进行显微镜下脑组织微血管内白细胞聚集和附壁计数。结果大鼠脑组织NF-κB活性和TNF-α蛋白水平以及脑组织含水量和脑组织微血管内白细胞聚集和附壁计数结果,模型组与假手术对照组比较均显著增高(P<0.05),七叶皂苷干预组与模型组比较均显著降低(P<0.05)。结论七叶皂苷可以抑制重症急性胰腺炎大鼠模型脑组织NF-κB活化,下调TNF-α蛋白的表达,减轻脑组织水肿程度,减少脑组织中白细胞在微血管壁的聚集和附着,从而对重症胰腺炎大鼠脑组织损害起到保护作用。 Objective To study the protective effects of aescine on the damage of brain tissue after severe acute pancreatitis in rats. Methods A total of 72 male Wistar rats were randomly divided into the sham operation group (normal control group), severe acute pancreatitis group and aescine treated group. Severe acute pancreatitis model in rats were induced by retrograde injection of 5% sodium taurocholate into the pancreatobiliary duct. A series of brain samples were obtained at 3, 6 and 12 hours respectively after operation in three groups. The activities of NF-κB activation of the brain were determined with immunohistochemistry and the levels of TNF-α protein in rat brain were measured with radioimmunoassay (RIA). In the meantime, the water content and accumulation of leucocytes and adhesion of rat brain were measured. Results Compared with sham operation group, NF-κB activities, the levels of TNF-α protein, the water content and leucocytes accumulation and adhesion of the rat brain were sig- nificantly increased (P〈0.05). Compared with SAP group,NF-κB activation,the levels of TNF-α protein,the water content and leucocytes accumulation and adhesion of the rat brain (P〈0.05) began to decrease obviously in aescine treated group (P〈0.05). Conclusion Aescine can dramatically inhibit NF-κB activities and TNF-α protein expression in the rat brain and alleviate rat brain edema, decrease leucocytes accumulation and adhesion of the rat brain, thus plays a protective role for the rat brain tissue damage after severe acute pancreatitis.
