摘要
目的:研究猪骨髓间充质干细胞(MSC)成骨分化(DOC)后在干扰素-γ(IFN-γ)刺激下对猪白细胞表面抗原(SLA)的变化及外周血单个核细胞(PBMC)增殖的影响,了解其免疫原性。方法:以未分化的MSC为对照,采用流式细胞术分别检测未诱导、成骨诱导的MSC、MSC+IFN-γ、DOC+IFN-γ的SLA分子的表达,采用单向混合淋巴反应观察各组细胞对外源凝集素(PHA)刺激PBMC的每分钟脉冲值(CPM)变化。结果:MSC+IFN-γ组和DOC+IFN-γ组SLA-I表达上调(P<0.05),SLA-Ⅱ表达明显上调(P<0.01)。MSC成骨分化后能抑制PHA刺激的PBMC增殖反应。结论:在体外实验中,DOC后的MSC在IFN-γ刺激下仍具有低免疫原性,且具有免疫调节作用。
Objective : To explore the immunogenicity of differentiated osteogenic ceils (DOC) from mesenchymal stem ceils (MSC). Methods:The expression of swine lymphocyte antigen (SLA) was separately determined by fluorescence -activated cell sorting in cell groups of MSC, MSC + IFN-γ, DOC and DOC + IFN-γ The phytohemagglutinin (PHA) induced proliferation of peripheral blood mononuclear cells (PBMC) was determined by one way mixed lymphocyte culturing with cells from the above groups. Results: SLA-Ⅰ expression was up-regulated in MSC + IFN-γ group and DOC + IFN-γ group ( P 〈 0. 05 ) , SLA- Ⅱ was up-regulated significantly (P 〈 0.01 ), DOC can suppress PHA induced PBMC proliferation. Con- clusion: Treated with IFN-γ or not, DOC possess low immunogenicity and perform immunogenic function in vitro.
