摘要
对肥胖病患者胰岛素与受体结合特性进行研究。采用放射受体技术对20例女性肥胖病患者和20例对照者的单个核细胞和红细胞与胰岛素结合进行测定。结果显示:1.对照组单个核细胞和红细胞与^(125)Ⅰ-胰岛素的最大特异结合分别为0.8%±0.06%(1.2×10~7细胞/ml)和13.5%±1.1%(3×10~9细胞/ml);而肥胖病组是0.42%±0.05%和9.1%±3.0%,均显示存在显著性差别,P分别<0.05和<0.001。2.肥胖病组红细胞胰岛素受体竞争抑制曲线和Scatchard图分析说明,受体特异结合降低主要由于受体数减少,亲和力在两组间并无明显差别,和3.血清胰岛素水平升高和胰岛素受体数降低存在显著负相关(r=—0.684,P<0.05)。
The study of characteristics of insulin binding to its receptor in obese patients was' presented. Using radioreceptor technique,insulin binding to monocytes and erythrocytes was measured in 20 female obese patients and 20 controls. The results showed that: (1)The percentage of maximal specific binding of 125I-insulin to monocytes and erythrocytes was 0. 8± 0. 06%(1.2× 107 cells/ml)and 13. 5 ±1. 1%(3×109 cell/ml Respectively in control group,whereas in obesity group the percentage was 0. 42 ± 0. 05% and 9. 1 ± 1. 3% ,P<0. 05 and <0. 001 respectively; (2)The competition curve and Scatchard plot analysis of insulin binding to erythrocytes from obese patients demonstrated that the specific binding reduction mainly due to a reduced number of receptors,although they did not show a significant differences in receptor affinity between the obesity group and controls;and (3)A negative correlation between the serum insulin level increase and insulin receptor binding sites reduction (P< 0.05).
