摘要
目的探讨高糖对体外培养兔视网膜Müller细胞表达血管内皮生长因子(VEGF)的影响。方法体外培养兔视网膜Müller细胞,采用免疫细胞化学方法半定量检测不同浓度葡萄糖对视网膜Müller细胞表达VEGF的影响。结果高糖可以明显增加Müller细胞VEGF的表达,且具有一定的浓度依赖性。结论高糖促进体外培养兔视网膜Müller细胞VEGF的表达,提示高血糖作为糖尿病视网膜病变(DR)的始动因素,其可能机制之一为诱导Müller细胞VEGF的表达。
Objective To investigate the effects of high level of glucose on expression of vascular endothelial growth factor (VEGF) in rabbit retinal Müller cells in vitro. Methods The cultured rabbit retinal Müller cells were divided into 6 groups,then incubated with 6 different concentration glucose respectively. VEGF expression in Müller cells was observed by immunocytochemistry (ICC). Results Comparing with groups of low level glucose, VEGF expression in cultured rabbit retinal Müller cells was significantly enhanced in high level groups, and this effect was concentration dependent in a certain range. Conclusion High level of glucose could increase VEGF expression in rabbit retinal Müller cells in vitro, which indicates that hyperglycaemia may promote the progression of diabetic retinopathy (DR) through the induction of VEGF expression in Müller cells.
