摘要 目的观察Cedemex对自然戒断吗啡依赖大鼠的内啡肽含量和多巴胺含量的影响。方法以剂量递增法连续皮下注射吗啡7d,建立吗啡依赖大鼠模型。用放射免疫法分别测定大鼠下丘脑和垂体中β-内啡肽(β-EP)的含量;用高效液相-荧光检测法测定大鼠伏隔核多巴胺的含量。结果(1)与停用吗啡组比较,Cedemex中、高剂量组能够显著升高下丘脑、垂体的β-EP水平(P〈0.05)。其中Cedemex中剂量分别升高的平均百分数为50%、印%;高剂量分别升高的平均百分数为55%、36%;(2)与停用吗啡组比较,Cedemex中、高剂量组能够显著降低吗啡戒断大鼠伏隔核的多巴胺水平(P〈0.05),降低的平均百分数分别为10%、17%。结论机体内啡肽和多巴胺的改变与吗啡依赖和戒断症状有关,Cedemex能够促进机体内啡肽和多巴胺水平的恢复。 Objective To observe the effects of Cedemex on contents of β-ndorphin(β-P) and dopamine in morphine dependent rats in the conditions of natural withdrawal. Methods A physical morphine dependent model of rats was established by subcutaneous injection of morphine in gradually increasing dosage within 7 days. The content of β-P in hypothalamus and pituitary in morphine dependent rats was detected using radio-immunoassay, and the content of dopamine in nucleus accumbens septi was detected using HPLC ( high performance liquid chromatography). Results ( 1 ) The contents of β-P and dopamine were significantly higher in Cedemextreated rats than those in the control (P〈0.05). They were raised respectively by 50% and 60% in middle dose of Cedemex-treated group, and raised respectively by 55% and 36% in high dose of Cedemex-treated group. (2) The content of dopamine in middle dose and high dose of Cedemex-treated group was significantly lower than that in the control ( P〈 0.05). The average percentages in both groups were reduced respectively by 10% and 17%. Conclusion The changes of the neurotransmitters such as β-P and dopamine are related with the withdrawal syndrome of the morphine dependent rats, and Cedemex can return the contents of β-P and dopamine to the normal levels.
