摘要 目的:探讨转移抑制基因Kail/CD82、ME491/CD63和MRP-1/CD9在卵巢癌发生、发展中的作用。方法:应用HIS方法检测30例正常卵巢组织、10例卵巢良性上皮肿瘤组织、6例卵巢交界性肿瘤组织和48例卵巢癌组织中Kail/CD82、ME491/CD63及MRP-1/CD9基因mRNA和蛋白的表达,及其在卵巢癌发生、发展中的作用和相关性,分析与临床病理因素的相关性。结果:Kail/CD82、ME491/CD63和MRP-1/CD9在卵巢癌组的年龄(P值分别为0·507、0·507和0·507)、病理分型(P值分别为0·071、0·070和0·072)上差异无统计学意义;高、中分化的表达与低分化比较差异有统计学意义,P值分别为0·038、0·038和0·037;在Ⅲ、Ⅳ期中表达减弱,与Ⅰ、Ⅱ期相比差异有统计学意义,P值分别为0·012、0·011和0·012;有、无淋巴转移的组织中的表达差异有统计学意义,P值分别为0·020、0·019和0·020。无淋巴转移者出现高表达。结论:Kail/CD82、ME491/CD63和MRP-1/CD9基因在卵巢癌中表达趋势一致,并随期别增加其mR-NA及蛋白表达逐渐降低,与卵巢癌的发生、发展负相关。 OBJECTIVE: To explore the effection of metastasis suppressor gene Kail/CD82, ME491/CD63 and MRP-1/CD9 in the occurrence and progressing of ovarian cancer. METHODS: The expression levels of Kail/CD82, ME491/CD63 and MRP-1/CD9 gene rnRNA and proteins were evaluated by using hybridization in site in normal ovarian tissues (n=30), ovarian benign tumor tissues (n=10), ovarian borderline tumor tissues (n=6) and ovarian cancer tissues (n=48). Analyze the expression levels of Kail/CD82, ME491/CD63 and MRP-1/CD9 genes and proteins by multivariant analysis statistically. Explore their effects on the invasion and metastases of ovarian cancer. To study the correlation of patients age at surgery, histological differentiation, lymph metastases, clinical grading and pathological typing. RESULTS: Kail/CD82, ME491/CD63 and MRP-1/CD9 were no statistically significant relationship between gene expression and the age at surgery (P=0. 507,0. 507, 0. 507) or clinical-pathological staging (P=0. 071,0. 070,0. 072). There was significant difference between G1 - G2 and G3 (P = 0. 038, 0. 038, 0. 037). We found obvious low expression levels in Ⅳ group (Ⅲ, Ⅳ period). Significant differences were noted between expression levels in Ⅲ, Ⅳ period and in Ⅰ,Ⅱ period,P=0. 012, 0. 011, 0. 012. There was significant difference between expression levels in tissues with lymph metastases and tissues without lymph metastases, P=0. 020, 0. 019, 0. 020. There was high expression in tissues without lymph metastases. CONCLUSIONS: The genes expression tendency of ME491/CD63, MRP-1/CD9 and Kail/CD82 in ovarian cancer is coincidence. Their mRNA and protein expressions gradually decrease with period raised and are negatively related with the occurrence and progressing of ovarian cancer.
