银杏叶片对结肠炎大鼠结肠组织IL-8表达的影响

摘要 目的观察银杏叶片(EGB)对结肠炎大鼠结肠组织IL-8表达的影响。方法利用2,4,6-三硝基苯磺酸(TNBS)灌肠制备大鼠实验性结肠炎模型。动物随机分为正常对照组、TNBS模型组、阳性药物对照组(5-ASA组,100mg/kg)和EGB组(200mg/kg)4组。评估大鼠结肠黏膜大体形态和组织学评分。生化法检测肠组织MPO和GSH-Px活性。采用ELISA法测定肠组织IL-8浓度。结果EGB能增加肠组织GSH-Px活性,降低MPO的活性,抑制IL-8的表达。结论EGB能减轻肠组织过量IL-8表达对实验性结肠炎大鼠的炎症损伤。 Objective To study the influence of Ginkgo biloba extract(EGB) on the expression of IL-8 in experimental colitis in rats. Methods Colitis in rats was induced by colonic administration with 2,4,6-trinitrobenzene sulfonic acid. The animals were divided into 4 groups:normal group,model group,5-ASA (100 mg/ kg) group,EGB (200mg/kg) group. The macroscopical and histological changes of the colon were evaluated. The activity of glutathion peroxide (GSH-Px) and myeloperoxidasea (MPO) in the colonic mucosa were determined. The IL-8 produced by colonic mulosal was analyzed with ELISA. Results As compared with model group, EGB could obviously increase the activity of GSH-Px, decrease the activity of MPO and the content of IL-8 in colon tissue. Conclusion EGB could protect colitis in rats from the inflammatory injury caused by IL-8.
