摘要
目的观察绿色荧光蛋白(GFP)转基因骨髓基质干细胞(BMSCs)移植至致痫鼠后的存活、迁移及其对癫痫鼠脑电的影响。方法分离、培养GFP转基因小鼠BMSCs,移植至青霉素致痫鼠的右侧海马内,比较移植后1w、2w、4wBMSCs在脑内的存活和迁移情况及大鼠脑电改变。结果BMSCs可以在致痫鼠脑内存活和迁移,随移植时间延长,细胞存活数逐渐减少(P<0.01);BMSCs移植可减少癫痫大鼠脑电的痫性放电,降低癫痫波波幅。结论BMSCs移植于青霉素诱发的癫痫鼠脑内后能够存活、迁移,并能够改善癫痫鼠的脑电生理功能,提示干细胞移植可能成为一种有效的癫痫治疗手段。
Objective To explore the survival and migration of bone marrow stromal ceils (BMSCs) of green fluorescent protein (GFP) transgenic mice in brain of epileptic rats and to investigate the effect of transplantation on the electroencephalogram of epileptic rats. Methods Bone marrow stromal cells of GFP were isolated and cultured in vitro, and then were transplanted into the right hippocampus of the penicillin induced epileptic rats. After transplantation for 1 w, 2 w, and 4 w, fluorescence microscope was used to observe the survival of transplanted bone marrow stremal cells and electrcencephalogram was used to monitor electrical activity of brains of the rats. Results It was found that the transplanted bone marrow stromal cells could survive and migrate in the brains of epileptic rats, and the number of the survival cells decreased gradually as the time went on (P〈0.01). Meanwhile the transplantation of stem cells could dramatically alleviate seizure activity in EEG recordings and reduce the amplitude of discharge. Conclusion It is found that bone marrow stromal cells transplanted into the hippocampus of the rat models of epilepsy can survive, migrate, and dramatically alleviate seizure activity in EEG recordings. It suggests that stem cell grafting may be a valid means for the cure epilepsy.
